Deep-Focus Diagnostics of Sunspot Structure
In sequel to Moradi et al. [2009, ApJ, 690, L72], we employ two established
numerical forward models (a 3D ideal MHD solver and MHD ray theory) in
conjunction with time-distance helioseismology to probe the lateral extent of
wave-speed perturbations produced in regions of strong, near-surface magnetic
fields. We continue our comparisons of forward modeling approaches by extending
our previous surface-focused travel-time measurements with a common midpoint
deep-focusing scheme that avoids the use of oscillation signals within the
sunspot region. The idea is to also test MHD ray theory for possible

On the nonthermal X-ray emission in blazar jets
We consider particle acceleration to high energy via diffusive shock
acceleration in a simple, self-consistent shock in jet model for blazars.
Electrons are assumed to be accelerated at a shock front in relativistic jets
and radiate synchrotron emission in a post-shock region. The full time, space
and momentum dependence of the electron distribution function is used for a
calculation of the nonthermal synchrotron spectra. We discuss the evolution of
the spectral index by varying the rate at which particles enter the
acceleration process. The results indicate that the synchrotron spectral index
displays a characteristic looplike behaviour with intensity (as has been
observed in several blazars), where the orientation of the loop depends on
whether the acceleration time scale is comparable to the synchrotron cooling
time scale or not. We show that our model provides a good fit to the observed

First report of venereal and vertical transmission of canine leishmaniosis from naturally infected dogs in Germany
<p>Abstract</p> <p>Background</p> <p>Canine leishmaniosis (CanL) is a zoonotic disease caused by <it>Leishmania (L.) infantum</it>. It is endemic to several tropical and subtropical countries but also to the Mediterranean region. It is transmitted by phlebotomine sandflies but occasional non-vector transmissions have been reported, including vertical and horizontal transmission.</p> <p>Findings</p> <p>The authors report a case of CanL in a female boxer dog from Dusseldorf, Germany, that had never been in an endemic region. A serum sample from the bitch was tested positive for antibodies against <it>Leishmania </it>(IFAT 1:2,000, ELISA 72). The bitch had whelped three litters, and one puppy from the third litter was also found to be seropositive for <it>Leishmania </it>antibodies (IFAT 1:4,000, ELISA 78).</p> <p>Conclusions</p> <p>Up to now, despite intensive searching, the occurrence of sandflies could not be proved in the bitch's region of origin. Thus, vertical and horizontal transmission are to be discussed as possible ways of infection. This may be the first report of venereal and vertical transmission of <it>L. infantum </it>in naturally infected dogs in Germany.</p
European Sea Bass (Dicentrarchus labrax) immune status and disease resistance are impaired by arginine dietary supplementation
Infectious diseases and fish feeds management are probably the major expenses in the aquaculture business. Hence, it is a priority to define sustainable strategies which simultaneously avoid therapeutic procedures and reinforce fish immunity. Currently, one preferred approach is the use of immunostimulants which can be supplemented to the fish diets. Arginine is a versatile amino acid with important mechanisms closely related to the immune response. Aiming at finding out how arginine affects the innate immune status or improve disease resistance of European seabass (Dicentrarchus labrax) against vibriosis, fish were fed two arginine-supplemented diets (1% and 2% arginine supplementation). A third diet meeting arginine requirement level for seabass served as control diet. Following 15 or 29 days of feeding, fish were sampled for blood, spleen and gut to assess cell-mediated immune parameters and immune-related gene expression. At the same time, fish from each dietary group were challenged against Vibrio anguillarum and survival was monitored. Cell-mediated immune parameters such as the extracellular superoxide and nitric oxide decreased in fish fed arginine-supplemented diets. Interleukins and immune-cell marker transcripts were down-regulated by the highest supplementation level. Disease resistance data were in accordance with a generally depressed immune status, with increased susceptibility to vibriosis in fish fed arginine supplemented diets. Altogether, these results suggest a general inhibitory effect of arginine on the immune defences and disease resistance of European seabass. Magnetic Coupling in the Quiet Solar Atmosphere
Three kinds of magnetic couplings in the quiet solar atmosphere are
highlighted and discussed, all fundamentally connected to the Lorentz force.
First the coupling of the convecting and overshooting fluid in the surface
layers of the Sun with the magnetic field. Here, the plasma motion provides the
dominant force, which shapes the magnetic field and drives the surface dynamo.
Progress in the understanding of the horizontal magnetic field is summarized
and discussed. Second, the coupling between acoustic waves and the magnetic
field, in particular the phenomenon of wave conversion and wave refraction. It
is described how measurements of wave travel times in the atmosphere can
provide information about the topography of the wave conversion zone, i.e., the
surface of equal Alfv\'en and sound speed. In quiet regions, this surface
separates a highly dynamic magnetic field with fast moving magnetosonic waves
and shocks around and above it from the more slowly evolving field of high-beta
plasma below it. Third, the magnetic field also couples to the radiation field,
which leads to radiative flux channeling and increased anisotropy in the
radiation field. It is shown how faculae can be understood in terms of this
effect. The article starts with an introduction to the magnetic field of the
quiet Sun in the light of new results from the Hinode space observatory and
with a brief survey of measurements of the turbulent magnetic field with the

Identifying factors likely to influence compliance with diagnostic imaging guideline recommendations for spine disorders among chiropractors in North America: a focus group study using the Theoretical Domains Framework
Background: The Theoretical Domains Framework (TDF) was developed to investigate determinants of specific clinical behaviors and inform the design of interventions to change professional behavior. This framework was used to explore the beliefs of chiropractors in an American Provider Network and two Canadian provinces about their adherence to evidence-based recommendations for spine radiography for uncomplicated back pain. The primary objective of the study was to identify chiropractorsβ beliefs about managing uncomplicated back pain without xrays and to explore barriers and facilitators to implementing evidence-based recommendations on lumbar spine xrays. A secondary objective was to compare chiropractors in the United States and Canada on their beliefs regarding the use of spine x-rays.
Methods: Six focus groups exploring beliefs about managing back pain without x-rays were conducted with a purposive sample. The interview guide was based upon the TDF. Focus groups were digitally recorded, transcribed verbatim, and analyzed by two independent assessors using thematic content analysis based on the TDF.
Results: Five domains were identified as likely relevant. Key beliefs within these domains included the following: conflicting comments about the potential consequences of not ordering x-rays (risk of missing a pathology, avoiding adverse treatment effects, risks of litigation, determining the treatment plan, and using x-ray-driven techniques contrasted with perceived benefits of minimizing patient radiation exposure and reducing costs; beliefs about consequences); beliefs regarding professional autonomy, professional credibility, lack of standardization, and agreement with guidelines widely varied (social/professional role & identity); the influence of formal training, colleagues, and patients also appeared to be important factors (social influences); conflicting comments regarding levels of confidence and comfort in managing patients without x-rays (belief about capabilities); and guideline awareness and agreements (knowledge).
Conclusions: Chiropractorsβ use of diagnostic imaging appears to be influenced by a number of factors. Five key domains may be important considering the presence of conflicting beliefs, evidence of strong beliefs likely to impact the behavior of interest, and high frequency of beliefs. The results will inform the development of a theorybased survey to help identify potential targets for behavioral-change strategies
Resistance of Leishmania (Leishmania) amazonensis and Leishmania (Viannia) braziliensis to nitric oxide correlates with disease severity in Tegumentary Leishmaniasis
BACKGROUND: Nitric oxide (NO(β’)) plays a pivotal role as a leishmanicidal agent in mouse macrophages. NO(β’ )resistant Escherichia coli and Mycobacterium tuberculosis have been associated with a severe outcome of these diseases. METHODS: In this study we evaluated the in vitro toxicity of nitric oxide for the promastigote stages of Leishmania (Viannia) braziliensis and Leishmania (Leishmania) amazonensis parasites, and the infectivity of the amastigote stage for human macrophages. Parasites were isolated from patients with cutaneous, mucosal or disseminated leishmaniasis, and NO(β’ )resistance was correlated with clinical presentation. RESULTS: Seventeen isolates of L. (L.) amazonensis or L. (V.) braziliensis promastigotes were killed by up to 8 mM of more of NaNO(2 )(pH 5.0) and therefore were defined as nitric oxide-susceptible. In contrast, eleven isolates that survived exposure to 16 mM NaNO(2 )were defined as nitric oxide-resistant. Patients infected with nitric oxide-resistant Leishmania had significantly larger lesions than patients infected with nitric oxide-susceptible isolates. Furthermore, nitric oxide-resistant L. (L.) amazonensis and L. (V.) braziliensis multiplied significantly better in human macrophages than nitric oxide-susceptible isolates. CONCLUSION: These data suggest that nitric oxide-resistance of Leishmania isolates confers a survival benefit for the parasites inside the macrophage, and possibly exacerbates the clinical course of human leishmaniasis
Variable selection for large p small n regression models with incomplete data: Mapping QTL with epistases
<p>Abstract</p> <p>Background</p> <p>Identifying quantitative trait loci (QTL) for both additive and epistatic effects raises the statistical issue of selecting variables from a large number of candidates using a small number of observations. Missing trait and/or marker values prevent one from directly applying the classical model selection criteria such as Akaike's information criterion (AIC) and Bayesian information criterion (BIC).</p> <p>Results</p> <p>We propose a two-step Bayesian variable selection method which deals with the sparse parameter space and the small sample size issues. The regression coefficient priors are flexible enough to incorporate the characteristic of "large <it>p </it>small <it>n</it>" data. Specifically, sparseness and possible asymmetry of the significant coefficients are dealt with by developing a Gibbs sampling algorithm to stochastically search through low-dimensional subspaces for significant variables. The superior performance of the approach is demonstrated via simulation study. We also applied it to real QTL mapping datasets.</p> <p>Conclusion</p> <p>The two-step procedure coupled with Bayesian classification offers flexibility in modeling "large p small n" data, especially for the sparse and asymmetric parameter space. This approach can be extended to other settings characterized by high dimension and low sample size.</p
Therapeutic and immunomodulatory activities of short-course treatment of murine visceral leishmaniasis with KALSOMEβ’10, a new liposomal amphotericin B
Visceral leishmaniasis (VL), a potentially fatal disease, is most prevalent in the Indian subcontinent,
East Africa and South America. Since the conventional antileishmanial drugs have many limitations we evaluated a
new ergosterol rich liposomal amphotericin B formulation, KALSOMEβ’10 for its leishmanicidal efficacy, tolerability
and immunomodulatory activity. Normal healthy mice were treated with 3.5 mg/kg single and 7.5 mg/kg single and double doses ofKALSOMEβ’10. Liver and kidney function tests were performed fourteen days after treatment. Next, normal mice were infected with Leishmania donovani amastigotes. Two months post infection they were treated with the above
mentioned doses of KALSOMEβ’10 and sacrificed one month after treatment for estimation of parasite burden in
the liver and spleen by Limiting Dilution Assay. Leishmanial antigen stimulated splenocyte culture supernatants were collected for cytokine detection through ELISA. Flow cytometric studies were performed on normal animals treated with KALSOMEβ’10, Amphotericin B (AmB) and AmBiosome to compare their immunomodulatory activities.
The drug was found to induce no hepato- or nephrotoxicities at the studied doses. Moreover, at all doses,
it led to significant reduction in parasite burden in two month infected BALB/c mice, with 7.5 mg/kg double dose
resulting in almost complete clearance of parasites from both liver and spleen. Interestingly, the drug at 7.5 mg/kg
double dose could almost completely inhibit the secretion of disease promoting cytokines, IL-10 and TGFΞ², and
significantly elevate the levels of IFNΞ³ and IL-12, cytokines required for control of the disease. Mice treated with KALSOMEβ’10 showed elevated levels of IFNΞ³ and suppressed IL-10 secretion from both CD4+ and CD8+ subsets
of T cells, as well as from culture supernatants of splenocytes, compared to that of normal, AmB and AmBisome
treated animal Treatment of infected mice with 7.5 mg/kg double dose of KALSOMEβ’10 was safe and effective in
clearing the parasites from the sites of infection. The drug maintains the inherent immunomodulatory activities of
AmB by effectively suppressing disease promoting cytokines IL-10 and TGFΞ², thereby boosting IL-12 and IFNΞ³ levels.
This emphasizes KALSOMEβ’10 as a promising drug alternative for lifelong protection from VL
